depends on what you are looking for and what your future plans are. 4-2-1 will give the most power in the middle rpm band range where most driving is done.
4-1 gives the most power at the top end rpms, ideal for racing. someone post the link for the great header test please.

I've heard that 4-1 gives more top end while 4-2-1 gives more low/mid. It's all about how your going to drive the car. You can try a search and you'll come up with pros/cons
