Default Vacuum question

I am making some itbs and just had a quick question, if I had a vacuum tube coming out after the throttle plates, will it still be getting vacuum all the time, even if the throttle plates are closed ?

after meaning closer to the head or farther. to answer your ? though. think of where all of the vacuum sources are in a regular manifold. between the throttle plate and head.
