Default Rear Balancer Shaft

Hi

I just wondering if it's a must to put a 6x100mm bolt into the rear balancer maintenance hole when I install the balancer belt, provided I had my own markings on the sprockets to align the belt right?
I got misfire after I replaced my timing belt stuffs. So I want to know if the rear balancer shaft is an issue.
If it's out of alignment, the engine just run roughly; but not missing, right?
PLEASE HELP! Thanks in advanced.

It's not absolutly necessary but it is much more accurate.
