You will have to remove the side mirrors and probably the inner door panel also. The piece is held on by a screw thats hidden under the mirror, and the rest is held on by small white plastic clips down the length of it, I think there is like 5 or 6. You also might have to remove the window... both cars I've removed the piece on already had the glass removed so I'm not sure if you can get to the clips with it in place. To get the clips out I pried them out of the door with a small screw driver, but you have to be very careful because the clips are pretty brittle after sitting in a door for 20 years. I broke a couple on each side but was able to find spares at a junk yard. The trim piece is made of a thin metal so you need to be gentle when pulling it out, they are very easy to bend out of shape. I ruined the first one I pulled out. Good luck
If you did a really good job of masking then I'm sure you could paint them on the car, but it would be a lot easier to get a good result if you took them off and sanded them off the car to remove all the old coating. You can probably remove them without breaking them if you are careful and have the right tools. I was in a junkyard and only had a few tools on hand so my situation was not ideal, and only broke a few of the clips.
I did it on my hatch. I did not want to remove them so I just stripped the coating and painted them..
Jasco paint stripper softens the rubber very well. Just be very carefull not to get it on your paint.
