Tuning a car for gas mileage
I've had a couple ideas. I'm running a 30% ethanol mix in my 1992 Celica GT right now, with no issues, and I've only lost about 1-2 mpg, but I save 40-50 cents a gallon. If I remember right, E85 will combust over a wider range of AFRs than gas will. One person I talked to has an E85 tune with a lean cruise equal to 16:1 on gas, I think it's 12:1 (E85).
Lean burn is a way to get better mileage, but you'll probably have higher NOx emissions, if they test for that in your area. Combustion chamber design is important to make sure what little fuel there is mixed properly.
Other than that, just make sure the ignition is in good order, the O2 sensor is working right (well everything that allows it to go to closed loop), and you can run skinnier tires with the correct pressure to save gas.
