No, you better do it when the car is on, thats always the smart thing to do.
That works even better on memory cards and jump drives, when you yank them out right as theyre saving. Try it, you will be amazed!

To clear codes and whatnot I always just pull my ecu fuse out for a 10-15 seconds... Less trouble than disconnecting the battery.
