I have a p28 w/ hondata s300 and I want to put this in. My question is do the s300s have preset settings (for a/f ratio, fuel mapping, etc.) that I can select from or do I have to set them myself? Would it hurt my engine to run with it for now w/out having to tune it? I'm gonna tune it later after I have all the parts installed.
I believe that there are settings for common combinations. I would just street tune it if you have the time. Just set your tune on the rich side and you should be fine until you can get the rest of your parts installed along with a dyno tune.

Yes... but in my opinion auto tune is a waste of time. It doesn't automatically set itself to the settings you want. It takes more time than a click of the button to calibrate everything.
You have a couple of maps to choose from that should get you up and running if you are running stock injectors. If you have anything other than stock as far as injectors or upgrades then you will need to modify your fueling and ignition tables accordingly.
