So what might this noise be when on the throttle?

For the past couple of years I've been hearing this low-pitched noise when on the throttle, mainly in 3rd-5th gears, mainly between 2K-4K rpm. It's kind of a low-pitched wawawawawawawawawa, and goes away as soon as I close the throttle.
It seems like the frequency of the "wah's" changes with speed. I guess that pretty much means it's transmission-related, right?
Car drives and shifts fine, always has. It's about to turn 300K miles on the original engine and transmission, but like I said everything feels fine when driving, still can get around 30 mpg average on my daily commute to and from work.
Any suggestions would be greatly appreciated.

i can tell you this much, anything that changes with speed noise wise is either suspension or tranny related, what exactly the sound is, i have no idea. Alot of times its difficult to diagnose problems until something actually breaks.
Gear whine?
The description sounds similar to what passengers try and describe the differential whine in my Spitfire as sounding like, which has about 1/8" too much clearance and needs a shim stuffed in there.
Check transaxle backlash perhaps?

Honestly, I've had "gear whine" from excessive backlash in my Spitfire for 15 years now. It isn't the end of the world, it just isn't as quiet or smooth as it used to be. Well, it isn't the end of the world until a gear tooth gets sheared off anyway...
Is the car somewhat "jerky" in first gear if you coast, press the gas, then go back to coast? I know, poor wording and a terrible way to try and describe backlash, but your description makes me think you should definitely look into the transmission for the source.

It's a little jerky at certain very low speeds in 1st gear if I do that, yeah, but it's always been like that since I bought the car 10 years ago. I've only noticed this sound in the past 2 years or less.
