Capabilities of the S2000 Trans./R.E.
So what all would it take to mod it to handle up to 600 lb/ft torque (hypothetically speaking so please don't flame me; its just a simple question)

As for the rear end, it can die @ stock power, or last well past that...all depends.
Prices vary, a BUILT tranny is $$, even the rear end can land you in the 3k range.
For a solid tranny/rear end, look to spend 10k JUST on the drivetrain.
for the diff, you can do 1 of 3 things that i know of
1. upgrade the stock differential with the comptech reinforcement. its fairly cheap ($400-500), but im not sure how well it will hold up to really high hp
2. nissan rear diff conversion. inlinepro runs this setup in their 9-second drag s2000, so it can hold some serious power. this option isnt cheap, i would assume its upwards of $1200
3. puttymod diff (i think thats how its spelled). i havent read up on these alot, but apparently they are very high quality and very durable. again, i would assume this option is expensive

600ft/lb of torque is probably going to be 900+whp which is a hell of a lot. I don't think even the drag S2k's run that much power.
for tranny options you can reinforce it but a regular OEM tranny with helical cut gears won't last very long at those power levels. you can cryotreat them or w/e i still don't think it'll last at 600ft/lb with a super stiff clutch grabbing it. IMO at 600ft/lb the only option is a PPG straight-cut gear'd tranny.
the diff I think the best option is to do a conversion either the R200 (its ~2500$ iirc through ILP) or one of the other custom ones like the ford rear end. puddymod, comptech reinforced, and OEM all have one problem. Honda made them. Although I love hondas and their associated parts, diff's definitely are not their forte. Puddymod I believe boasts 3x the original toughness or something along those lines, but you're shooting for 4x the torque and it won't be anywhere near close to a gradual transition. i'd imagine with 600ft/lb your power delivery's gonna be crap downlow and violent up top.

i had an 05 s2k, bought it brand new, i had an act heavy duty clutch setup, clutch slave mod , cai and exahust and gave my car hell on the track and street. Did several redline launches at the track and never had any problems with the trans or rear end.
ran 13.40s all day long and i had 50k miles on her when i sold her and still was good, i always heard the rear ends were weak but never happened to me.

yeah... im pushing the 1200 marker... in order to do so, im gonna piggyback 2 ebay electric superchargers on the intake and see where im at...
The OEM trans, regardless of how you drive it, will eventually let go at anything over 600whp. I've broke 2 in one month, just putting my foot to the floor rolling in 2nd gear. The stock rear end, I have no personal experience with, as my car has had an R200 in it ever since it was turbo'd.
Keep it under 600whp, and shift sensibly, and it'll last a long time.
