Default Spark plug

I'm trying to trouble shoot something and I need to have my my spark plugs not fire one at a time to figure it out. Can I have my car running and just remove the spark plugs one at a time? Or does the car need to be off, then remove the spark plug, then start the car?

Well, for future reference to anyone. You can. By that I mean, remove the boots as the car is running.

Yes you can, it's called a cylinder drop test.
